What a 3-part analyser reports

Cells are counted and sized by impedance. White cells are sorted by size into lymphocytes, mid-sized cells (monocytes, eosinophils and basophils together) and granulocytes. With the red-cell and platelet results, that typically comes to about 19–21 parameters from 10–20 µL of EDTA whole blood, at around 40–60 samples an hour on a compact model.

The three reagents

Diluent

Dilutes the sample and carries cells through the counting aperture; also used to rinse the fluid path between samples.

Lyse

Breaks open red cells so white cells can be counted and haemoglobin measured. Most modern lysing reagents are cyanide-free.

Cleaner

Removes protein build-up from the aperture and tubing, usually at shutdown and during periodic maintenance.

Keeping cost per test down

  • Choose reagent pack sizes you will finish within their opened stability at your volume.
  • Follow the manufacturer’s advice on standby versus full shutdown, so you are not paying for unnecessary start-up cycles.
  • Train at least two staff so samples are not sent out when one is on leave.
  • Record reagent lot numbers and run controls after each lot change to avoid repeat testing.

Frequently asked questions

What does the “mid” cell group mean on a 3-part analyser?
It is the group of medium-sized white cells — mainly monocytes, eosinophils and basophils counted together. A 3-part analyser cannot report them separately.
How long do 3-part reagents last once opened?
It depends on the product; the manufacturer states the opened stability on the label or insert. Choose pack sizes you will use up within that period at your daily volume.
Can a 3-part analyser run finger-prick samples?
Many compact 3-part analysers have a pre-diluted mode for capillary samples, which helps with children. Check that the model you are offered supports it.
